Thanks very much, Murray Singer Murray Singer, Professor Department of Psychology University of Manitoba Winnipeg, Manitoba Canada R3T 2N2 Ph: 204-474-8486 murray.singer(a)umanitoba.ca Title: The Representation of Episodic Memory in Single Neurons of the Human Hippocampus Time and place: Thurs, Oct. 17, 2013, 3 PM (snacks at 2:30); Department of Psychology, University of Manitoba, P412 Duff Roblin The hippocampus is known to play a fundamental role in declarative memory, but it is not known how mnemonic information is coded by the activity of individual hippocampal neurons. Three coding schemes have been considered: a localist coding scheme (i.e., "grandmother" cells), a fully distributed coding scheme, and a sparse distributed coding scheme. Sparse distributed coding has long been hypothesized to be the most efficient way for hippocampal neurons to rapidly encode pattern-separated episodic memories without overwriting previously stored information. We measured the activity of hippocampal neurons in 9 epilepsy patients undergoing intracranial monitoring as they completed a recognition memory test. Single-unit activity was higher in response to targets (previously studied words) compared to foils (novel words). This effect occurred because, for each neuron, only a few targets elicited a strong response, as uniquely predicted by the longstanding theory that episodic memory is supported by a sparse distributed neural code. John Wixted is an acclaimed leader in research concerning the basic character of human memory, applications such as eyewitness memory, and the brain mechanisms of memory function. He regularly offers groundbreaking advances by challenging and often refuting existing theory and analysis. In this regard, he recently discovered that people's recollection of learning contexts makes a continuous contribution to memory performance, in contrast with the previously-held all-or-none view. Likewise, he has collaboratively provided evidence that a contemporary view of the role of the hippocampus in recognition-memory appreciably oversimplifies the true state of affairs. Dr. Wixted achieves these advances by means of strikingly insightful, simple, and elegant experimental design. In his investigations of the continuous versus discrete nature of contextual recollection, he examined the distributions of people's ratings of their recollective experiences on scales such as 1-20 or 1-100. This approach, which oddly had escaped other investigators, clarified the memory processes under scrutiny. Dr. Wixted has an outstanding capacity to make his material accessible and fascinating to the listening audience, almost regardless of the inherent interest value of the lecture topic. He simplifies complex issues and may even leave the audience feeling sorry that the talk is over. Dr. Wixted's contributions have earned him many recognitions and awards during his career. He served as editor of the Psychonomic Bulletin & Review from 1998 to 2002. He is a Fellow of the prestigious Society of Experimental Psychologists. In 2011, that organization awarded him the Howard Crosby Warren Medal, previously conferred on individuals including Karl Lashley and B. F. Skinner.

Manitoba Neuroscience Network Seminar Friday, May 31, 2013 | 9:00 am Dr. Gordon Glazner Associate Professor - Department of Pharmacology & Therapeutics, University of Manitoba & Principal Investigator, Division of Neurodegenerative Disorders, St. Boniface Hospital Research Topic: Investigating the role of NF-kB in diabetic neuropathy. Location: PX236/238, Psychiatry Bldg. Bannatyne Campus In people with diabetes, a major complication is neuropathy, in which sensory nerve endings die back from the limbs. This leads to severe pain, numbness, skin ulcerations and poor would healing, and often ends in amputation of the limb. We and others have found that NF-kB, a protein that regulates the production of other proteins, is decreased in the sensory nerves of diabetic animals. We've also found that both the hormones that stimulate NF-kB and a key antioxidant protein produced by NF-kB action are decreased in sensory neurons. We propose now to use our new knowledge to find if activation of this pathway can inhibit diabetic neuropathy, and to find out if loss of this pathway in diabetes is the root cause of neuropathy. This project is separated into five separate aims. In Aim 1 we will deliver a NF-kB stimulating hormone, called CNTF, to diabetic animals, and we will determine if this can reduce or reverse formation of diabetic neuropathy. In Aim 2, we will see if inhibition of this pathway, either by specific drugs or by genetic deficiency, can increase the severity and risk for development of diabetic neuropathy in rodents. In Aim 3, we will investigate whether diabetes inhibits more members of the newly discovered pathway, and if we can rescue neurons by reversing this loss. In Aim 4 we will discover whether increasing anti-oxidant action can prevent formation of diabetic neuropathy. In aim 5, we will examine what it is about diabetes that causes inhibition of this pathway. Our group has recently discovered a pathway that normally keeps sensory nerves healthy is disrupted in diabetes. We believe that this disruption is the underlying cause of neuropathy. The proposed work will take these findings into whole animal "proof of concept" studies to determine if manipulation of this pathway can be used therapeutically, and will help to discover the underlying cause of diabetic neuropathy. Subject: Manitoba Psychological Society Spring Workshop on DSM-V On Friday, June 7, 2013, the Manitoba Psychological Society is holding its 2013 Spring Institute titled Developments in Mental Disorder Diagnosis: DSM-IV, DSM-V, and the Road Ahead. This day long workshop featuring Dr. Robert Krueger, a Hathaway Distinguished Professor in the Department of Psychology at the University of Minnesota and a member of the DSM-V Personality and Personality Disorder Working Group, will address the following learning objectives: 1. To understand major aspects of the history of psychiatric classification that led to the approach taken in DSM-IV. 2. To understand notable changes between DSM-IV and DSM-V, such as the removal of the multiaxial system. 3. To understand both the DSM-IV (copied in DSM-V Section II) and DSM-V Section III approaches to classifying personality disorders, as well as the conceptual underpinnings of these approaches. 4. To understand notable limitations of the DSM paradigm, current efforts to develop more empirically-based alternatives (e.g., the NIMH RDoC initiative), and the reasons why new empirically-based approaches are needed. Our dynamic and inspiring speaker, Dr. Esther M. Sternberg is internationally recognized for her discoveries in brain-immune interactions and the effects of the brain’s stress response on health and illness: the science of the mind-body connection. Physician and Medical Researcher, Dr. Sternberg’s discoveries have revealed a complicated working relationship between the senses, the emotions, and the immune system. She has found that “the brain’s hormonal response to stress plays a role in the body’s susceptibility to autoimmune inflammatory disease, and to how severe it becomes”. Dr. Sternberg received her MD and training in Rheumatology at McGill University. She was on faculty at Washington University in St. Louis, Mo, (1980-86); Senior Scientist in the National Institutes of Health in Maryland (1986-2012) and is Research Professor at American University in Washington, D.C. She recently joined the University of Arizona’s Center for Integrative Medicine as the first Director of Research at the Integrative Medicine Center in Tucson. Author of numerous original scientific and review articles published in leading scientific journals including Nature Medicine, Science, New England Journal of Medicine, Scientific American, J. Multi-user Brain Canada funding opportunity - please help

Brain Canada will soon request applications for Platform Support Grants (PSGs) that are, "intended to contribute to the operating costs of national or regional research platforms that serve large numbers of researchers in the neurosciences". The draft request for applications for this initiative (www.braincanada.ca/files/Draft_RFA_for_PSGs_March_2013.pdf) makes it clear that it will be essential to improve, upgrade, link and coordinate existing infrastructure so that it works better and/or is more widely accessible, ultimately accelerating neuroscience research. The intention is to fund 10 grants at up to $1 million pa for 3 years. Interested stakeholders, including researchers and funding partners, met on April 12th and decided that the Manitoba neuroscience community is in a position of strength to be active in this competition. We intend to build an application that emphasizes our human brain imaging capabilities. We have a nationally distinct portfolio of human neuroimaging approaches, including Magnetic Resonance Imaging (MRI), PET, CT, intraoperative MRI and soon combined PET/MRI. We also have a new complementary core with strength in small animal PET, MRI, CT, IVIS and 2-photon imaging, and numerous in vitro imaging capabilities spanning several department and institutes. Supporting all of these are core facilities in epigenetics, proteomics, experimental radiopharmacy and others. It will be our position that we can bind our imaging modalities together as a uniquely innovative offering of approaches to facilitate neuroscience research from bench to bedside. At least, this will be presented as a Provincial platform in neuroimaging, and it remains possible with further development that the application could be for a prairie-wide regional platform. The innovative and translational nature of the Kleysen Institute will be a cornerstone of this strategy. At this stage, it is imperative to get a strong sense of the potential users of this neuroimaging platform. I am therefore asking you to send me a short paragraph describing whether any of the imaging techniques mentioned above do or will serve your research interests, and how. Please add anything in particular you would need in terms of platform support to facilitate your work in these neuroimaging directions more efficiently. Would you use any of the techniques if you didn't have to operate the equipment yourself, but had an individual dedicated to its operation for researchers? Some have said that providing technical expertise would make for easier accessibility. Others have mentioned that employing a strong computational biologist would facilitate image analysis that would otherwise be next to impossible. Please be creative. This is your chance! You are not making any commitments and you need not be detailed. You may also speculate on how the platform might be useful to you even if you do not have concrete plans at this point. We are looking for as many responses as we can get and ultimately, the success of this effort is predicated on having the largest group of impacted users possible. Thank you for your attention. We use 2-photon imaging of brain slices to test neuron-astrocyte communication in neurovascular coupling. We plan to use this technique also in live animals in the small animal imaging core. Using discoveries about neurovascular coupling in vitro and in vivo, we would like to manipulate blood flow to improve regional brain metabolism after injury using PET in animals and eventually in humans. PET in animals and humans is an area of particular interest for me. There is much potential in using new PET probes to study different aspects of brain function. We have a unique capability to synthesize new PET probes and I am interested in having a radiochemist employed to link this expertise to research in PET imaging of brain function after injury. Biosketch: Dr. Rastegar is an Assistant Professor at the Regenerative Medicine Program and the Department of Biochemistry and Medical Genetics at the University of Manitoba. She is currently a scientist member of Manitoba Institute of Child Health (MICH) and Molecular Genetics and Human Disease Group (MGHDG). Dr. Rastegar completed her PhD in Biomedical Sciences at the "Université Catholique de Louvain (UCL)", Belgium. She continued her research training in US and Canada. In Canada, Dr. Rastegar performed her postdoctoral training at the McGill Cancer Centre in Montreal and at the Hospital for Sick Children in Toronto. Dr. Rastegar is a widely recognized researcher in the field of epigenetics in brain development and neurological disorders. Her current research is focused on the role of MeCP2 as an epigenetic factor and modeling neurological diseases in brain-derived neural stem cells. Biosketch: Dr. Rastegar is an Assistant Professor at the Regenerative Medicine Program and the Department of Biochemistry and Medical Genetics at the University of Manitoba. She is currently a scientist member of Manitoba Institute of Child Health (MICH) and Molecular Genetics and Human Disease Group (MGHDG). Dr. Rastegar completed her PhD in Biomedical Sciences at the "Université Catholique de Louvain (UCL)", Belgium. She continued her research training in US and Canada. In Canada, Dr. Rastegar performed her postdoctoral training at the McGill Cancer Centre in Montreal and at the Hospital for Sick Children in Toronto. Dr. Rastegar is a widely recognized researcher in the field of epigenetics in brain development and neurological disorders. Her current research is focused on the role of MeCP2 as an epigenetic factor and modeling neurological diseases in brain-derived neural stem cells. 2013 Nick Shepel Student Travel Award Competition deadline: May 10th, 2013 The Nick Shepel Student Travel Award was established by the Division of Neurodegenerative Disorders, St. Boniface Hospital Research to honor Nick Shepel who was an outstanding researcher, friend and leader. Nick was a true inspiration to his lab partners and brought fire, humor, and a desire to always do better. His insistence to always try better will have a permanent effect on his lab mates. Nick led by example and taught that detailed preparation and execution of experiments ensured reliable results. Purpose: The purpose of this fund shall be to recognize a young investigator (graduate student within the Faculty of Medicine - University of Manitoba) by providing an annual award (valued at up to $1,000) to support travel costs associated with the applicant's attendance at a national/international conference where the applicant is presenting a poster or oral presentation. Eligibility: At the time of application, students must be registered in the Faculty of Medicine at the University of Manitoba. 1. "Registered" includes being accepted by the major department of study, admitted by the Faculty of Graduate Studies, or eligible to register in the Faculty of Graduate Studies. 2. Dr. Li is currently the Director of the Neuroscience Research Program at the Faculty of Medicine, and holds a cross appointment in the Department of Human Anatomy and Cell Science. Dr. Li received his M.D. and PhD in Physiology at Norman Bethune University of Medical Sciences in Changchun, China and completed his residency in Psychiatry at University of Saskatchewan. Dr. Li completed postdoctoral training in the Department of Psychiatry at University of Saskatchewan and at Harvard University. Dr. Li began his academic career at University of Saskatchewan as Assistant Professor (1992) and was Director of the Neuropsychiatric Research Unit prior to moving to University of Manitoba in 2007. Dr. Li's research interests are primarily biological psychiatry. His research uses animal models to examine the role of white matter and neurodegeneration in mood disorders and schizophrenia using cellular, molecular, microMRI imaging, and behavioural assessment techniques. He also studies the mechanisms of treatment in psychiatric and neurodegenerative diseases, focusing on the neuroprotective effects of antidepressants and atypical antipsychotics. His work includes research collaborations in North America and China. Dr. Li has received research support from Canadian Institute of Health Research (CIHR), Manitoba Health Research Council (MHRC), Canadian Psychiatric Research Foundation (CPRF), Alzheimer's Society of Canada, Schizophrenia Societies, as well as the pharmaceutical industry. Dr. Wafa Kammouni Research Associate, Department of Internal Medicine, University of Manitoba Topic "Exploring oxidative stress induced by rabies virus in dorsal root ganglia." Dr. Wafa Kammouni is a Research Associate in the Department of Internal Medicine under the supervision of Dr. Alan Jackson. He is studying rabies virus interactions with dorsal root ganglia. He has developed primary cultures of dorsal root ganglia since they are permissive to infection and resistant to apoptosis. He has also studied oxidative stress-induced by rabies virus infection in vitro. By applying immunofluorescence techniques and western blotting Dr. Kammouni pointed out that oxidative stress response occurs in cultured dorsal root ganglia, that this phenomenon is controlled by anti-oxidants and that there is increased amount of nuclear factor kB (NF-kB) without an increase in the activity leading to a possible defect in anti-oxidant response. Manitoba Neuroscience Network Seminar Series Friday, February 22nd, 2013 | 2:00 pm Dr. Derek van der Kooy Professor Department of Molecular Genetics University of Toronto Topic: Where Brains Come From Location: Theatre B Derek van der Kooy served as Professor in the Department of Anatomy and Cell Biology at the University of Toronto from 1991 until 2002, when he became a Professor in the Department of Molecular Genetics. Derek received a M.Sc. in Psychology at the University of British Columbia, and a Ph.D in Anatomy, first at Erasmus University in the Netherlands, and finishing in the Department of Anatomy at the University of Toronto. Dr. van der Kooy gained postdoctoral research experience at Cambridge University in England and at the Salk Institute in California. The van der Kooy lab works on various stem cell biology and developmental biology research projects; specifically, stem cells in organisms from Drosophila to humans. We produced the first report of stem cells in the adult mammalian eye, published in 2000 in Science. We also have isolated a rare stem cell from the adult mouse and human pancreas that can show extensive proliferation under defined conditions in vitro. Of interest to the lab is the lineage of neural stem cells from pluripotent embryonic stem cells, with relevance to the origin of the earliest neural stem cell in the developing embryo. ABSTRACT: One of the few neurobiological facts not anticipated by Cajal is the existence of neural stem cells in the embryonic and adult mammalian brain that can produce new neurons in the adult brain (new neurons first shown by Altman). The earliest mammalian neural stem cells differentiate from pluripotent embryonic stem cells. These primitive neural stem cells emerge in response to LIF and have a wider non-neural phenotypic potential than later neural stem cells. By embryonic day 8.5 in mouse, true FGF2 dependent neural stem cells emerge and by embryonic day 14 they have given rise to copies of themselves and to EGF dependent neural stem cells. The separate FGF2 and EGF dependent neural stem cells increase greatly in numbers later in neurogenesis, but by E14 appear identical to adult neural stem cells. The adult mammalian neural stem cells are mostly quiescent, dividing asymmetrically only once every few weeks. Mouse embryonic stem (ES) cells cultured in low cell density, completely defined media adopt a neural identity. Using a clonal colony-forming assay, we identify the novel primitive neural stem cell stage as a component of neural lineage specification (this cell is similar to the one we isolate from the early embryo), which is negatively regulated by TGFb-related signaling. These results are consistent with a default mechanism for neural fate specification. Primitive neural stem cells are formed directly from single ES cells in a LIF-dependent manner, express multiple neural precursor markers and give rise to neurons and glia. Moreover, in vivo mouse chimera experiments reveal that these primitive ES-derived neural stem cells have a broad range of neural and non-neural lineage potential. These results support a model whereby definitive neural stem cell formation is preceded by a primitive neural stem cell stage during neural lineage commitment. Most recently, we have found that the LIF-dependent, primitive neural stem cells persist in adult organisms and can repopulate a depleted definitive neural stem cell pool in the adult. Garfield Weston Foundation ------------------------------------ I am writing as the Executive Director, Neuroscience of The W. Garfield Weston Foundation. The Foundation is a private Canadian family foundation where donations are directed to qualified donees in the fields of education, land conservation, science in Canada's North and through unsolicited Trustee Initiated Grants. Last year, the Foundation launched a pilot funding program for neuroscience research within the Greater Toronto Area. I am excited to let you know the Foundation is launching another pilot funding program for neuroscience research this week open to institutions across Canada. We wanted to ensure you and the researchers at the University of Manitoba were aware of this opportunity. The goal of this request for proposals (RFP) is to provide funding for translational research or clinical trials that will have transformational impact in neurodegenerative diseases of aging. The Foundation will commit up to $4 million to this program. Grants will be for up to $500,000 per year for up to three years to a maximum of $1.5 million per project. Application requires a one to two page Letter of Intent due Monday, March 25th, 2013. High potential applicants will be invited to submit a short proposal, and grantees will be announced in the summer. Dear All, As you may know, The Manitoba Neuroscience Network will be hosting Dr. Derek van der Kooy on Feb 22, 2013. He is currently a Professor at the Department of Medical Genetics. Dr. van der Kooy is one of the pioneers of the field of adult stem cells. His publications in the field of Neural stem cells, Retinal stem cells and pancreatic stem cells are published in top journals such as Cell, Science, Nature, Neuron,Development and J Neuroscience are highly cited. Dr. van der Kooy is also a great speaker and I will hope that you will try to attend his lecture on Feb 22, at noon, Room 626 in Regenerative Medicine. I am trying to schedule his short day visit on Feb 22nd and therefore I am asking those interested in meeting him to contact me at eftekhar(a)cc.umanitoba.ca. Manitoba Neuroscience Network Seminar Series Friday, February 22nd, 2013 | 12:00 noon Dr. Derek van der Kooy Professor Department of Molecular Genetics University of Toronto Topic: Where Brains Come From Location: Regenerative Medicine Lecture Theatre (626 BMSB) Derek van der Kooy served as Professor in the Department of Anatomy and Cell Biology at the University of Toronto from 1991 until 2002, when he became a Professor in the Department of Molecular Genetics. Derek received a M.Sc. in Psychology at the University of British Columbia, and a Ph.D in Anatomy, first at Erasmus University in the Netherlands, and finishing in the Department of Anatomy at the University of Toronto. Dr. van der Kooy gained postdoctoral research experience at Cambridge University in England and at the Salk Institute in California. The van der Kooy lab works on various stem cell biology and developmental biology research projects; specifically, stem cells in organisms from Drosophila to humans. We produced the first report of stem cells in the adult mammalian eye, published in 2000 in Science. We also have isolated a rare stem cell from the adult mouse and human pancreas that can show extensive proliferation under defined condi-tions in vitro. Of interest to the lab is the lineage of neural stem cells from pluripotent embryonic stem cells, with relevance to the origin of the earliest neural stem cell in the developing embryo. ABSTRACT: One of the few neurobiological facts not anticipated by Cajal is the existence of neural stem cells in the embryonic and adult mammalian brain that can produce new neurons in the adult brain (new neurons first shown by Altman). The earliest mammalian neural stem cells differentiate from pluripotent embryonic stem cells. These primitive neural stem cells emerge in response to LIF and have a wider non-neural phenotypic potential than later neural stem cells. By embryonic day 8.5 in mouse, true FGF2 dependent neural stem cells emerge and by embryonic day 14 they have given rise to copies of themselves and to EGF dependent neural stem cells. The separate FGF2 and EGF dependent neural stem cells increase greatly in numbers later in neurogenesis, but by E14 appear identical to adult neural stem cells. The adult mammalian neural stem cells are mostly quiescent, dividing asymmetrically only once every few weeks. Mouse embryonic stem (ES) cells cultured in low cell density, completely defined media adopt a neural identity. Using a clonal colony-forming assay, we identify the novel primitive neural stem cell stage as a component of neural lineage specification (this cell is similar to the one we isolate from the early embryo), which is negatively regulated by TGFb-related signaling. These results are consistent with a default mechanism for neural fate specification. Primitive neural stem cells are formed directly from single ES cells in a LIF-dependent manner, express multiple neural precursor markers and give rise to neurons and glia. Moreover, in vivo mouse chimera experiments reveal that these primitive ES-derived neural stem cells have a broad range of neural and non-neural lineage potential. These results support a model whereby definitive neural stem cell formation is preceded by a primitive neural stem cell stage during neural lineage commitment. Most recently, we have found that the LIF-dependent, primitive neural stem cells persist in adult organisms and can repopulate a depleted definitive neural stem cell pool in the adult.
