Reminder: Manitoba Neuroscience Network Seminar Series Friday, January 18th, 2013 | 12:00 noon Dr. Samuel David Professor - Department of Neurology & Neurosur-gery, Centre for Research in Neuroscience, Research Institute of the McGill University Health Centre Topic: Targeting inflammation in spinal cord injury Location: Theatre B, Bannatyne Campus Short Biosketch: Dr. Sam David is a Professor in the Department of Neurology and Neurosurgery, at the Centre for Research in Neuroscience, The Research Institute of the McGill University Health Center in Montreal. He obtained his PhD degree in 1979 from the University of Manitoba, and did postdoctoral work at McGill University and University College London, UK. His research focuses on molecular mechanisms of inflammation after spinal cord and peripheral nerve injury, CNS autoimmune disease, and the molecular control of iron homeostasis in the CNS and its dysregulation in CNS pathologies. He has published in top tier journals such as Science, PNAS, Neuron, Nature Immunology, Nature Reviews Neuroscience, Brain, and Journal of Neuroscience. He is Director of the CIHR Training Program in Neuroinflammation, which includes 16 laboratories at three Montreal area Universities. He is the current President of the Canadian Association of Neuroscience, member of the Medical Advisory Committee of the MS Society of Canada, member of the Scientific Advisory Board of Wings for Life Spinal Cord Research Foundation, and other organizations. After a short visiting scholarship in Baylor College of Medicine, he did three postdoctoral fellowships in University of Massachusetts and University of Manitoba. He joined the Department of Human Anatomy and Cell Science, University of Manitoba as an Assistant Professor in 2002. He was promoted to Associate Professor in 2006 and Professor in 2011. Dr. Kong's research program is directed to the understanding of molecular regulation of neuronal cell death in neurodegenerative diseases. Using in vivo and in vitro models of stroke, amyotrophic lateral sclerosis, Alzheim-er's disease, schizophrenia and depression, his group examines posttranslational oxidative modification of SOD1 as a factor of age-related onset of neurodegenerative diseases, investi-gates the role of mitochondrial death-inducing proteins in regulating mitophagy and delayed neuronal death in stroke, and tests a demyelinating hypothesis of schizophrenia. Dr. Kong is a recipient of the prestigious Heart and Stroke Foundation of Canada New Investigator award (2004-2009) and a principal investigator of the Canadian Stroke Network (since 2007), which is one of the Centers of Excellence of Canada. His research program is continuously support-ed by international and national grant agencies such as the Muscular Dystrophy Association (USA), National Natural Science Foundation of China, Canadian Institutes of Health Research, Canadian Stroke Network, Canada Foundation for Innovation, and Heart and Stroke Foun-dation of Canada. Microglia are the principal immune-response cells in the central nervous system. Resting microglia constantly survey the microenvironment in the normal brain. Upon brain dysfunction, microglia are activated and exert detrimental or beneficial effects on the surrounding neurons. Microglia have remarkably fast and dynamic activities in both the normal and pathological brain. An exciting possibility is that they are communicating with neurons through ion channel mechanisms. In neuronal circuits, microglia are assumed to be crucial to synaptic pruning and plasticity, yet no direct evidence has been identified. Microglia are strongly activated in pathological conditions such as pain, stroke and neurodegeneration, however, the molecular mechanisms for microglial activation and function in brain diseases are still controversial. Overall, we are interested in three interrelated topics on microglia-neuron communication: (1) The ion channel mechanism of microglia-neuron communication; (2) Microglia in synaptic function; and (3) Microglia's contribution to neuropathic pain. We have been exploring microglia chemotaxis (Wu et al., Glia, 2007, Fig.1), microglia in synaptic plasticity (Wu and Zhuo, J Neurophysiol, 2008), as well as an unique microglial voltage-gated proton channel, Hv1, in the brain (Wu et al., Nat Neurosci, 2012). Taking Hv1 proton channel as an entry point, our initial project will study direct communication from microglia to neurons through microglial Hv1. We will then explore how microglia function in neuronal circuits under normal and pain conditions. The results from our studies would advance the understanding of microglia function in the brain and provide therapeutic targets for neuropathic pain treatment. Recently large number of pharmaceutical companies pulled out of Neuroscience drug development research. This is likely to impact the field. Why did they leave? My research interest may have some explanations to offer. Behavioral motor effects can be studied using animals; but not the internal sensations of higher brain functions. Since understanding the latter is required to reverse its defects in most of the disorders of higher brain functions, studies in this direction may help to select appropriate pharmaceutical agents that can reverse the disease symptoms. I will focus my talk on some of the theoretical approaches that I have been taking to understand internal sensations. Research summary: I am interested in understanding how internal sensations are created from neuronal activities. Specific features of some of the diseases are likely to provide clues to understand the normal functioning of the nervous system from which formation of internal sensations may be understood. Short bio sketch: After graduating Medicine in 1988 and practicing family medicine for a short period, I did MD program in Biochemistry at the Calicut University, India. This was followed by a Research Associate position at the Jawaharlal Nehru University, New Delhi to study negative regulatory elements upstream of p53 gene. I moved to Canada in 1999, did MSc (under Dr.Umberto DeBoni) and PhD (under Dr. Min Zhuo) from the University of Toronto. Later, I did post-doctoral training in Dr. Mark Zylka's laboratory at the University of North Carolina, Chapel Hill. Currently, I am a 4th year Resident in Neurology at the University of Manitoba and am spending part time in Dr. Albensi's laboratory to identify a biomarker for stroke. Location: PZ236/238 Psychiatry Bldg., Bannatyne Campus Short Biosketch: Dr. Jun-Feng Wang recently joined the University of Manitoba, in March 2012, as Associate Professor in the Department of Pharmacology and Therapeutics and as an adjunct faculty member in the Department of Psychiatry. His research focuses on the role of oxidative stress and oxidative stress-induced protein modifications in bipolar disorder; and on understanding the neuroprotective mechanisms of mood stabilizing drugs. Dr. Wang received his Ph.D. in Neuroscience in 1991 from Peking University. He did his Post Doc training at University of California San Francisco and McMaster University. Dr. Wang received his first academic appointment as Assistant Professor at McMaster University from 2001 to 2003. Later he joined the Centre for Addiction and Mental Health and University of Toronto as a Research Scientist and Assistant Professor from 2003 to 2007. Dr. Wang was at in the Department of Psychiatry at University of British Columbia as Assistant Professor from 2007-2010, and as Associate Professor from 2010-2012. Dr. Wang has received considerable research support, including grants from the Canadian Institutes of Health Research, Ontario Mental Health Foundation, Stanley Foundation Research Grants, and the Canadian Psychiatric Research Foundation. He has twice received National Alliance for Research on Schizophrenia and Depression Young Investigator Awards. His research is currently supported by grants from the Manitoba Health Research Council, the Canadian Institutes of Health Research and University of Manitoba start-up funding. Indeed, identifying the physical basis of memory within the brain (the memory trace) has been a long-standing challenge for scientists since Karl Lashley's "search for the engram" in the 1950's. Memories are thought to be encoded by sparsely distributed groups of neurons. However, identifying the precise neurons supporting a given memory (the memory trace) has been a long-standing challenge. We have shown previously that lateral amygdala (LA) neurons with increased CREB are preferentially activated by fear memory expression, suggesting they are selectively recruited into the memory trace. Here we used an inducible diphtheria-toxin strategy to specifically ablate these neurons. Selectively deleting neurons overexpressing CREB (but not a similar portion of random LA neurons) after learning blocked expression of that fear memory. The resulting memory loss was robust and persistent, suggesting that the memory was permanently erased. These results establish a causal link between a specific neuronal subpopulation and memory expression, thereby identifying critical neurons within the memory trace. Short bio: The research in Dr. Josselyn's lab is dedicated to understanding the neural basis of cognitive function and dysfunction. To unravel the molecular, cellular and circuit processes that underlie learning and memory, her lab uses a multidisciplinary approach include the use of genetically-engineered mice, viral vectors, cellular imaging, electrophysiology and detailed behavioral analysis. Her program of research focuses on two main elements 1) examining the brain regions and molecules responsible for normal memory formation and 2) using this knowledge to intervene in conditions in which memory is impaired (for instance in neurodegenerative diseases such as Alzheimer's disease). She has published extensively in the top scientific journals on these subjects and has shown that fear memories in mice can be "erased" and is currently examining novel treatments for the memory disorders that characterize Alzheimer's disease. Dr. Davie is presently the Scientific Director of the Manitoba Health Research Council, Leader of the Terry Fox Research Institute Prairie Node, and Professor in the Department of Biochemistry and Medical Genetics at the University of Manitoba. He serves on several Editorial Boards of journals publishing in Biochemistry, Cell Biology and Molecular Biology and is Editor of the journal Biochemistry and Cell Biology. He has served as a Chair and panel member on CIHR and NCIC/CCSRI Peer Review Committees. His research interests include epigenetic regulation of gene expression in normal and disease (cancer) cells, nuclear matrix structure and function, sub-cellular trafficking of transcription factors and chromatin remodeling complexes, signal transduction pathways, chromatin structure and function, and biomarkers in the early detection of disease. He currently holds a Canada Research Chair in Chromatin Dynamics (Tier 1). He then went to the University of Alberta for a Post Doctoral Fellowship, which he completed in 1992. He then came to the University of Manitoba to assume the post of Assistant Professor. He rose through the ranks to become Professor in 2002. He became a Tier 1 Canada Research Chair in 2006. Hatch has won numerous career awards from the Manitoba Health Research Council, Heart and Stroke Foundation of Canada, Canadian Institutes of Health Research and the University of Manitoba, and he has published over 100 refereed articles, reviews and book chapters. Hatch relocated his laboratory to MICH in June 2011 and is looking forward to fruitful and productive collaborations with the MICH community. Studies on the regulation of phospholipid and lipid & lipoprotein metabolism with a focus on the childhood diseases Barth Syndrome & Persistant Pulmonary Hypertension of the Newborn. Dr. Hatch's laboratory is studying whether increasing the cells ability to remodel cardiolipin will aid in the treatment of Barth Syndrome, a rare genetic disease that weakens the heart. He and his colleagues are guided by the belief that their work may lead to new therapies to treat a range of diseases and disorders from Barth Syndrome to heart failure. A few years ago Hatch received a small grant from the Children's Hospital Foundation and subsequently a MICH grant to study Barth Syndrome. Based upon the foundation of these grants he published several papers on Barth Syndrome and was subsequently successful in obtaining several grants from the Barth Syndrome Foundation USA and most recently a five year grant from the Canadian Institutes of Health Research to continue with his studies. Indeed, identifying the physical basis of memory within the brain (the memory trace) has been a long-standing challenge for scientists since Karl Lashley's "search for the engram" in the 1950's. Memories are thought to be encoded by sparsely distributed groups of neurons. However, identifying the precise neurons supporting a given memory (the memory trace) has been a long-standing challenge. We have shown previously that lateral amygdala (LA) neurons with increased CREB are preferentially activated by fear memory expression, suggesting they are selectively recruited into the memory trace. Here we used an inducible diphtheria-toxin strategy to specifically ablate these neurons. Selectively deleting neurons overexpressing CREB (but not a similar portion of random LA neurons) after learning blocked expression of that fear memory. The resulting memory loss was robust and persistent, suggesting that the memory was permanently erased. These results establish a causal link between a specific neuronal subpopulation and memory expression, thereby identifying critical neurons within the memory trace. Short bio: The research in Dr. Josselyn's lab is dedicated to understanding the neural basis of cognitive function and dysfunction. To unravel the molecular, cellular and circuit processes that underlie learning and memory, her lab uses a multidisciplinary approach include the use of genetically-engineered mice, viral vectors, cellular imaging, electrophysiology and detailed behavioral analysis. Her program of research focuses on two main elements 1) examining the brain regions and molecules responsible for normal memory formation and 2) using this knowledge to intervene in conditions in which memory is impaired (for instance in neurodegenerative diseases such as Alzheimer's disease). She has published extensively in the top scientific journals on these subjects and has shown that fear memories in mice can be "erased" and is currently examining novel treatments for the memory disorders that characterize Alzheimer's disease. Teaching opportunity for senior graduate student

Dear Members and Non-members, In an effort to reach out to the undergraduate student population, the Department of Pharmacology and Therapeutics offers undergraduate Pharmacology courses at the Fort Garry Campus (PHAC 4030 in Fall and 4040 in Winter). There is a substantial neuroscience/neuropharmacology component to PHAC 4030 and I have traditionally offered a senior graduate student an opportunity to teach 2 basic neurochemistry and neuroanatomy lectures, with mentorship, in this course. I believe this could represent valuable teaching experience for graduate students and I have an opening this Fall (2012) in November. If you are a 3rd year or higher graduate student in a program with emphasis in neuroscience and would like to gain some teaching experience, please consult your graduate advisor first, and then reply to me for further discussion. It will be of particular interest to graduate students whose research is in Neuroscience (registration is open to all graduate students). This course will provide an introduction to the structure & function of the human central nervous system from an anatomical perspective. The course has 2 components: 1. Dear Neuroscience Principal Investigators, As you are aware, in 2010 the Manitoba Neuroscience Network joined forces with Clinical Neuroscience Rounds to present one basic neuroscience seminar per month during the second hour of Rounds on the last Friday of each month. The benefits are: · Dedicated basic neuroscience seminar series · Basic scientists and clinicians have a consistent opportunity for discussion once per month · Clinical neuroscience training program is exposed to basic science research In 2012/2013 I would like to continue our arrangement and expand the overall quality of the offering by ADDING 2 VISITING SPEAKER SLOTS out of the 7 total for the academic year. The Manitoba Neuroscience Network will fund these speakers at reasonable levels. I am looking for a volunteer FACULTY MEMBER to coordinate the series. Seminar Notice Monday June 11 2012 2.30pm TOPIC: Novel roles for netrin-1 and DCC regulating myelination and synaptic plasticity in the CNS Timothy E Kennedy, PhD Professor Dept of Neurology and Neurosurgery Montreal Neurological Institute McGill University Dr. Kennedy's laboratory is investigating the molecular mechanisms that direct neural development and how such mechanisms influence the organization and plasticity of the mature mammalian central nervous system. These studies aim to identify the biochemical signals that direct cells and axons to move, as well as the target-derived signals that instruct them to stop once they reach their goal and to make appropriate connections with neighboring cells. Indeed, identifying the physical basis of memory within the brain (the memory trace) has been a long-standing challenge for scientists since Karl Lashley's "search for the engram" in the 1950's. Memories are thought to be encoded by sparsely distributed groups of neurons. However, identifying the precise neurons supporting a given memory (the memory trace) has been a long-standing challenge. We have shown previously that lateral amygdala (LA) neurons with increased CREB are preferentially activated by fear memory expression, suggesting they are selectively recruited into the memory trace. Here we used an inducible diphtheria-toxin strategy to specifically ablate these neurons. Selectively deleting neurons overexpressing CREB (but not a similar portion of random LA neurons) after learning blocked expression of that fear memory. The resulting memory loss was robust and persistent, suggesting that the memory was permanently erased. These results establish a causal link between a specific neuronal subpopulation and memory expression, thereby identifying critical neurons within the memory trace. Short bio: The research in Dr. Josselyn's lab is dedicated to understanding the neural basis of cognitive function and dysfunction. To unravel the molecular, cellular and circuit processes that underlie learning and memory, her lab uses a multidisciplinary approach include the use of genetically-engineered mice, viral vectors, cellular imaging, electrophysiology and detailed behavioral analysis. Her program of research focuses on two main elements 1) examining the brain regions and molecules responsible for normal memory formation and 2) using this knowledge to intervene in conditions in which memory is impaired (for instance in neurodegenerative diseases such as Alzheimer's disease). She has published extensively in the top scientific journals on these subjects and has shown that fear memories in mice can be "erased" and is currently examining novel treatments for the memory disorders that characterize Alzheimer's disease. Topic: Novel mechanisms in the pathogenesis of diabetic retinopathy. Dysfunction of endothelial cell (ECs) causing increased production of vasoactive factors and extracellular matrix (ECM) proteins are characteristic features of chronic diabetic complications such as diabetic retinopathy. Glucose-induced biochemical alterations in the ECs activate a cascade of signaling pathways leading to such changes. Chronic diabetes leads to the activation of a number of signaling proteins including protein kinase C and advanced glycation end product formation. These signaling cascades are activated in response to hyperglycemia-induced oxidative stress. Such aberrant signaling leads to activation of transcription factors, such as nuclear factor-κB and activating protein-1. Although transcription factors are of importance in the regulation of protein production, they remain ineffective without transcriptional co-activators. In diabetes, transcriptional co-activator p300, with histone acetyl transferase activity, regulates several transcription factors. In addition, microRNA alterations regulate gene expression and the downstream effects eg. increased vasoactive factor and ECM protein production. Blockers of oxidative stress and histone acetylation as well as selected miRNA mimics prevent such alteration. Hence a complex web of pathways including intracellular signaling, epigenetics and miRNA alterations are involved in the pathogenesis of functional and structural changes in chronic diabetic complications. We have identified specific changes in the ECs and in the organs affected by diabetic complications such as retina. We have also identified novel adjunct treatment strategies, targeting epigenetic and transcriptional machinery for chronic diabetic complications such as retinopathy. Call for Proposals for 2012 Spinal Cord Injury Research Projects The Manitoba Spinal Cord Injury Research Committee (MSCIRC) is pleased to announce that the 2012 Grants and Awards competition is now underway and accepting applications for spinal cord injury research projects in Manitoba. Purpose Grants provided through this initiative aim to increase understanding of spinal cord function; reduce the disability related to spinal cord injury; increase functional capacity after spinal cord injury; reduce the incidence and effects of secondary complications after spinal cord injury; and increase quality of life and community integration after spinal cord injury. Eligibility Grant applications will only be considered from accredited educational institutions, disability related organizations, research institutions or members of hospital staff. Dear Manitoba Neuroscientists, The Manitoba Schools Science Symposium (MSSS) is being hosted at the University of Manitoba from April 26th to April 29th. This is an event that showcases the hard work of students in Manitoba at all levels, from elementary to high school. The WCSN will continue our involvement in the MSSS again this year by offering 3 project awards related to neuroscience. We feel that this is part of our contribution to growing the neuroscience discipline at the grassroots level in Manitoba. To make this happen, I am looking for volunteer project judges from within our membership to rank the top neuroscience-related projects. Projects will be displayed in the Max Bell Centre and judging will take place Saturday morning, April 28, from 9AM to Noon. Lunch will be provided after the judging is complete. CENTRE ON AGING PRESENTS: 2011-2012 Research Seminar Series Ben Albensi, PhD, Associate Professor Department of Pharmacology and Therapeutics Tuesday, April 10, 2012 Room 405 Brodie Centre 12:00 p.m. to 1:00 p.m. TOPIC: The Involvement of NF-kB Signaling in Memory Impairment in Alzheimer's Disease Memory impairments are central to a variety of neurological conditions, but the most familiar condition where memory is impaired is in Alzheimer's disease. Treating Alzheimer's disease pathology and the cognitive deficits that are seen in this condition remain problematic. Part of the challenge of course is that if we do not understand the fine details of how memory works on a basic biological or neurochemical level, then we are not able to effectively treat memory impairments. However, over the last several years research associated with the biological basis of memory has made great strides. We now believe that Alzheimer's disease is a disease of short term memory where the conversion of short term to long term memory becomes impaired. Recent developments also suggest that transcription is essential for the consolidation of long term memory. In addition, transcriptional signaling pathways such as NF-kB play not only a role in cancer and inflammatory responses, but also in long term memory and in Alzheimer's disease. The focus of this talk will be to discuss NF-kB signaling in memory and in Alzheimer's disease. In particular, new data from our lab show several novel links in the NF-kB pathway that suggest distinct roles for NF-kB's gene targets in differentially affecting memory and Alzheimer's disease pathology.
