Dr. Adrian Owen is a Canada Excellence Research Chair in Cognitive Neuroscience and Imaging from Western University (formerly UWO). He will be in Winnipeg for International Brain Awareness Week March 15 and 16, and will deliver a public lecture at 6:30 pm Thursday March 15 in the Frederic Gaspard Theatre at the Bannatyne Campus. Manitoba Neuroscience Network Seminar -Dr. Sari Hannila Date: Friday, Feb 24th, 2012 Time: 9:00 am Location: PX236/238 Psychiatry Bldg., Bannatyne Campus Dr. Sari Hannila Assistant Professor, Department of Human Anatomy & Cell Science Faculty of Medicine, University of Manitoba Topic: Downregulation of Smad2 by secretory leukocyte protease inhibitor enhances axonal regeneration in the CNS Dr. Hannila is originally from Sudbury, Ontario, and received her Bachelor of Science degree in Life Sciences from Queen's University in 1999. She then completed her PhD in the Department of Anatomy and Cell Biology at Queen's under the supervision of Dr. Michael Kawaja. Prior to joining the University of Manitoba in July, 2010, Dr. Hannila worked as a postdoctoral fellow in the laboratory of Dr. Marie T. Filbin at Hunter College in New York City. Her research focuses on the neurobiology of developing and regenerating axons in the central nervous system, with a particular interest in spinal cord injury. When the spinal cord is injured, regeneration of damaged axons is inhibited by CNS myelin and this leads to permanent paralysis. During her postdoctoral fellowship, Dr. Hannila discovered that a protein called secretory leukocyte protease inhibitor (SLPI) can reverse the inhibitory effects of myelin and enhance axonal growth. She is now building on these findings as an independent researcher at the University of Manitoba. She is currently testing SLPI in a rat model of spinal cord injury to determine if SLPI can promote axonal regeneration and improve functional recovery. If SLPI proves to be effective, she plans to conduct further studies in which SLPI will be administered in more complex, clinically-relevant models of spinal cord injury. In addition to its pro-regenerative effects, SLPI also has neuroprotective properties and to diversify her research program, Dr. Hannila plans to test SLPI in models of neurodegenerative diseases such as multiple sclerosis, amyotrophic lateral sclerosis, Alzheimer's, and Parkinson's disease. These studies will be complemented by ongoing experiments that will investigate the molecular mechanisms underlying SLPI's effects on axonal growth and neuronal survival, and this work will focus on SLPI's ability to regulate gene and protein expression. Research Interests: Pain sensitivity within the skull is restricted to the intracranial meninges, the system of membranes that envelops the brain. Afferent thinly myelinated Aδ-fibres and unmyelinated C-fibres that originate from the trigeminal ganglion densely innervate the meninges, in particular the cranial dura mater. It is widely recognized that activation or sensitization of these dural nociceptive afferents is responsible for the genesis of migraine headaches. However, any details of the pathophysiological mechanisms involved are largely speculative and remain to be discovered. Identification of the underlying mechanisms of activation of dural nociceptors may have important implications for understanding and mitigating the pathogenesis of migraine headaches. Research Interests: Pain sensitivity within the skull is restricted to the intracranial meninges, the system of membranes that envelops the brain. Afferent thinly myelinated A -fibres and unmyelin-ated C-fibres that originate from the trigeminal ganglion densely innervate the meninges, in particular the cranial dura mater. It is widely recognized that activation or sensitization of these dural nociceptive afferents is responsible for the genesis of migraine headaches. However, any details of the pathophysiological mecha-nisms involved are largely speculative and remain to be discovered. Identification of the underlying mechanisms of activation of dural nociceptors may have important implications for understanding and mitigating the pathogenesis of migraine headaches. Dear Manitoba Neuroscientists, We are now in the process of planning the 2012 Manitoba Neuroscience Annual Meeting. A date will be announced soon. I have planned the last 2 meeting programs myself and I am looking for fresh insight this year. I have received passing suggestions ranging from keeping the meeting format essentially the same, to changing to a trainee-centered event, to holding the meeting every 2 years for lack of new content, to having it planned as an overnight retreat to facilitate evening socialization. I want this to be the best event possible. I am therefore soliciting meeting format and location suggestions from potential participants. The WHO informs us that by 2025 there will be 300 million sufferers from diabetes worldwide - a figure approximately equal to the population of the USA. Currently close to 50% diabetic patients develop some form of peripheral nerve disease, which can lead to loss of protective sensation and limb amputation. The incidence is expected to rise 10-fold in the next 10 years in First Nations peoples of Canada. There is no treatment. My group is, therefore studying pathogenetic mechanisms and potential drug targets. Key approaches involve identification of the cellular transduction of damage signals (neurotrophic deficits, oxidative stress, glucose stress, mitochondrial dysfunction) and failure of cellular protective mechanisms. Techniques: cell culture, confocal microscopy, fluorescence imaging, neuronal plasmid transfection, general biochemistry and pharmacology. This year our feature event will be a scientific visit and public presentation by Dr. Adrian Owen from the University of Western Ontario on March 15 and 16. Dr. Owen was recently recruited to Canada from Cambridge University as a Canada Excellence Research Chair (CERC) in Cognitive Neuroscience and Imaging. He is coming at the invitation of the Winnipeg Chapter Society for Neuroscience, the Faculty of Medicine and the President of the University of Manitoba. Dr. Owen's itinerary will include a minisymposium on cognitive neuroscience and brain imaging, to showcase work being done in Manitoba, as well as an open scientific seminar and an evening public lecture. Manitoba Neuroscience Network Meeting Friday, November 25th, 2011 9:00 - 10:00am Dr. Larry Jordan Professor, Department of Physiology University of Manitoba Topic: Control of locomotion by a serotonergic pathway arising in the parapyramidal region of the medulla Location: PX236/238 PsychHealth, Bannatyne Campus Brief Bio: Born in Paris, Texas, in 1944, I grew up in Texarkana, Texas, the son of a bakery worker/farmer and a first grade teacher. I earned a B.A. degree in zoology at the University of Texas at Austin, and then entered graduate studies at The University of Texas Southwestern Medical School in Dallas, where I was supervised by William D. Willis, Jr., a student of John Eccles in Canberra, Australia, and a former President of the Society for Neuroscience. I moved to Winnipeg in 1970 to assume a post-doctoral position with Dr. John Phillis, also trained in Canberra, who was a pioneer in CNS neuropharmacology. John moved to Saskatoon to take up the position of Head of Physiology there, and I remained in Winnipeg, supported by a Medical Research Council Scholarship. Since that time I was able to participate in the creation of the Winnipeg Spinal Cord Research Centre (http://www.scrc.umanitoba.ca/), with funding from the Health Sciences Centre Research Foundation, the annual Will-to-Win Golf Tournament (2012 will be its 29th year!), the Canadian Institutes for Health Research, the International Human Frontier Science Program, the Canadian Network of Centres of Excellence, and the U.S. National Institutes of Health. I was the PI in the Canadian Institutes of Health Research Group on Spinal Cord Functional Systems for a number of years. I initiated the formation of the Winnipeg Chapter of the Society for Neuroscience, and I served two terms as Head of the Department of Physiology at the University of Manitoba (1992-2001). My research has focused on the neural control of locomotion, with progress along three major lines of investigation: (1) identification of the brain areas and neural pathways responsible for providing descending messages for the initiation of locomotion, (2) the study of spinal cord neurons that control locomotion at the spinal level and (3) discovery of new physiological properties of spinal motoneurons and interneurons that are central to their recruitment during locomotion. I am currently focusing on a descending pathway from a discrete serotonergic nucleus in the medulla that is involved in the initiation of locomotion and in control of inter- and intralimb coordination, and I am doing experiments to evaluate the potential of these neurons to restore coordinated locomotor movements after transplantation into the spinal cord of paraplegic rats. Seminar Series Dr. Julia Rempel Director, Liver Research Program Assistant Professor, Dept Medicine/Immunology University of Manitoba "Hepatitis C virus and interferon treatment induced neuropsychiatric disorders" Friday, November 04 12:00 - 13:00 Pharmacology Library - Room A229 Chown Building ** **Video-linked to St. Boniface Research Centre Room R1002 Bio: Dr. Julia Rempel received her PhD at The University of Manitoba in the Department of Immunology in 1999 (Kent HayGlass supervisor). She did her post-doctoral fellowship at the Scripps Research Institute, La Jolla, CA, examining how viruses can cause disease through activating deleterious pro-inflammatory immune responses in mouse models of encephalitis. During this time, reports emerged suggesting that despite a high burden of infectious disease within Aboriginal peoples, Aboriginal ancestry allowed for more effective clearance of hepatitis C virus infection. Intrigued, she returned to Manitoba in 2002 as a member of the Section of Hepatology to study the interaction of Aboriginal immunity and hepatitis C infection. Her laboratory (dubbed the OOH-QUIN Immunology Laboratory, OOH-QUIN means liver in Cree and Oji-cree) has been/is at the forefront in unveiling how immune genetics and function influences disease outcomes in Aboriginal populations. She has received national grants from CIHR and the Canadian Liver Foundation, as well as local and pharmaceutical funding. The trainees within her laboratory have also received numerous awards and fellowships. In addition to scientific publications, she has made extensive contributions to knowledge translation within the Aboriginal community, including the production of a research video in Oji-cree. Abstract of Research Interests: The cornerstones of Dr. Rempel's research program are innate immunity, liver disease, and Aboriginal health. The primary focus of her laboratory has been exploring immune processes in the enhanced ability of Aboriginal people's to spontaneously clear hepatitis C virus relative to other peoples. With a combination of genetic and cell behavior assays, her research team found that immune cells from Aboriginal peoples have a greater genetic tendency to pro-inflammatory immunity and a reduced susceptibility to hepatitis C virus protein deregulation relative to Caucasian population controls. These ongoing studies involve Aboriginal communities, as well as partners from the University of Manitoba, the National Microbiology Laboratory (NML) and inner city clinics. Clinical studies in collaboration with the Section of Hepatology and Viral Hepatitis Investigation Unit continue to evaluate immune responses of Aboriginal and Caucasian patients undergoing treatment for chronic hepatitis C infection. As part of these studies, they are considering the potential of baseline immunity to predict therapeutic clearance and neuropsychiatric side effects. More recently, her team has been concerned with the negative consequences of a highly pro-inflammatory immune environment within the Aboriginal population. To this end, they are working with the Pediatric Diabetes Unit to examine immunopathogenesis in type 2 diabetes of Aboriginal youth. MARK FRY Assistant Professor, Department of Biological Sciences University of Manitoba Topic: Roles of the subfornical organ in regulating energy balance. Location: PX236/238 PsychHealth, Bannatyne Campus The main focus of the research carried out in the lab is to understand the physiology of neurons that regulate homeostasis. In particular, the lab is interested in understanding regulation of electrical activity in neurons that control food intake and other aspects of energy homeostasis. Neurons of the sensory circumventricular organs (CVOs) are thought to be especially important for regulation of homeostasis because these neurons are not protected by the blood-brain barrier, and are able to sense hunger-stimulating and satiety (hunger-ending) hormones circulating in the bloodstream. A second focus of research aims to understand roles of different ion channels in the regulation of electrical excitability of neurons. Ion channels are proteins in neuronal cell membranes that undergo conformational changes to form a pore that allows charged ions such as Na+, Ca++, K+ and Cl- to flow into or out of the cell. These "ionic currents" are the basis of the neuronal action potential and neuronal communication within the CNS. There is a large diversity of ion channels: dozens of ion channel gene families have been described, and many families have numerous isoforms. These isoforms exhibit only subtle structural differences, but often exhibit dramatic functional differences. Populations of neurons may express vastly different complements of these isoforms and the relationship between ion channel isoform expression and the patterns of neuronal electrical activity is only beginning to be understood. Acutely modulating neuronal ion channel properties by activating intracellular signalling pathways or by changing expression patterns of ion channels by disease or other stimulus can result in dramatic changes in neuronal function. We focus on neurons that coordinate an organism's response to stress, with a particular interest in clarifying how the molecules released at the onset of a stressful stimulus leave a lasting imprint on how 'stress-relevant' circuitry functions. Within this context, we conduct experiments that will allow us to understand the fundamental rules that govern cell to cell communication within the hypothalamus and elucidate the molecular machinery that contributes to changes in synaptic function which, in turn, may be critical for changing network output.

Summary of Research: Dr. Brent Fedirchuk's Summary of Research: * Dr. Fedirchuk's research is aimed at determining the factors regulating the excitability of spinal neurons and determining how this regulation contributes to the production of useful motor output, such as locomotion. Using single-cell electrophysiological recording techniques coupled with experimental preparations able to exhibit complex motor activity, Dr. Fedirchuk and colleagues have been able to demonstrate a rapid and reversible enhancement of the excitability of spinal neurons from descending and intrinsic spinal systems. * Dr. Fedirchuk is a native Manitoban, and received a B.Sc. (Hons) in Zoology from the University of Manitoba, followed by M.Sc. and Ph.D. degrees working with Dr. Susan Shefchyk in the Department of Physiology (University of Manitoba). Dr. Fedirchuk then undertook postdoctoral training (CIHR Fellowship) with Dr. Hans Hultborn (Copenhagen) and Dr. Michael O'Donovan (NIH). Dr. Fedirchuk was recruited back to Winnipeg, initially as a Research Associate in Dr. Larry Jordan's laboratory, and subsequently as an Assistant Professor. Baxter Associate Professor Department of Neuroscience, Mount Sinai School of Medicine Abstract: Episodic memory depends on a network of interconnected brain structures. Damage to either the fornix or mammillary bodies causes amnesia in both humans and nonhuman primates. We have found that the severity of amnesia after damage to either of these structures is modulated by prior loss of cholinergic projections to the inferotemporal cortex, the cortical region through which visual information reaches these structures. Macaque monkeys with cholinergic depletion of inferotemporal cortex had unimpaired episodic memory performance. However, these monkeys were substantially more impaired following additional lesions of either the fornix or the mammillary bodies than were monkeys with intact temporal cortical cholinergic input. Reversing the order of the lesions resulted in less severe amnesia, indicating that the presence of cholinergic projections to temporal cortex at the time of fornix or mammillary body damage facilitates recovery. These findings show that humans with loss of cortical cholinergic function, for example in Alzheimer's disease, may be less able to adapt to the impairments caused by neuronal damage within cortical and subcortical structures important for memory. Consequently, drugs that increase cortical cholinergic neurotransmission may slow disease progress by augmenting this adaptive process. (Kinesiology/Anatomy & Physiology) Simon Fraser University Teaching Interests Neuroscience Cellular and Systems Level Physiology Research Interests Pain sensitivity within the skull is restricted to the intracranial meninges, the system of membranes that envelops the brain. Afferent thinly myelinated A��-fibres and unmyelinated C-fibres that originate from the trigeminal ganglion densely innervate the meninges, in particular the cranial dura mater. It is widely recognized that activation or sensitization of these dural nociceptive afferents is responsible for the genesis of migraine headaches. However, any details of the pathophysiological mechanisms involved are largely speculative and remain to be discovered. Identification of the underlying mechanisms of activation of dural nociceptors may have important implications for understanding and mitigating the pathogenesis of migraine headaches. We have developed a novel en bloc dural-skull preparation that allows us for the first time to apply optical imaging techniques to directly study with high resolution the free nerve endings and terminals of the nociceptive fibres that terminate in the dura mater. The objective of our research is to gain insight into the neurophysiology of peripheral pain activation to better understand the pathophysiological processes that occur during migraine headache. It is our hope that this will lead to new peripheral drug targets and offer additional therapeutic treatments for this devastating illness. Laboratory Techniques We are primarily an optical imaging laboratory that uses both two-photon laser scanning imaging as well as widefield epifluorescence microscopy. Microscopes are configured for both microfluorometric and transmitted light imaging using photomultiplier tube or high sensitivity EMCCD camera based detection. We combine optical imaging of calcium signaling selectively within individual terminals with electrophysiological stimulation, pharmacological manipulations, and UV and two-photon photolysis of caged compounds. Date: Friday, June 3, 2011 Time: 12:00 PM - 1:00 PM Location: Theatre C Dr. Michael Kawaja Professor, Department of Anatomy and Cell Biology Queen's University at Kingston Topic: Nerve Growth Factor - The driving force behind sympathetic sprouting. Biosketch: Dr. Kawaja did his undergraduate studies in biology at Acadia University and completed his doctoral degree in Anatomy at the University of Western Ontario in 1989. He then went on to have two MRC-funded post-doctoral positions, the first with Dr. Fred Gage at the University of California San Diego, and the second with Dr. John Roder at the Samuel Lunenfeld Institute at the Mount Sinai Hospital in Toronto. In 1993 Dr. Kawaja commenced his MRC-funded scholarship position as an Assistant Professor in the Department of Anatomy and Cell Biology at Queen's University. Over the last 17 years he has risen through the ranks of Associate and Full Professor at Queen's. Dr. Kawaja has held numerous operating grants from CIHR (MRC), Heart and Stroke Foundation, Ontario Neurotrauma Foundation, and Ontario Mental Ontario Ministry for Research and Innovation. He has served as a panel member of CIHR Scholarships and continues to serve as a panel member for CIHR Neuroscience "B". He has also served on a number of peer review panels including those for Alzheimer Society of Canada, Heart and Stroke Foundation of Canada, Multiple Sclerosis Society of Canada, Ontario Mental Health Foundation, and Ontario Ministry of Research and Innovation Early Researcher Awards. health Foundation. His research focuses on gaining a greater understanding of the reparative mechanisms in the mammalian central and peripheral nervous systems, as a consequence of injury and disease.
